Ferric Citrate

Ferric citrate is a reddish-brown, odorless, slightly hygroscopic powder consisting of ferric ions complexed with citrate anions. It is sparingly soluble in water and forms stable complexes in solution.

Chemical Name and CAS Registry Number: Iron(III) citrate [3522-50-7]

Functional Category: Iron supplement; phosphate binder; hematinic

Pharmacopeial Specifications:

  • Iron Content: 16.5–21%

  • Identification: Positive for iron and citrate

  • Loss on Drying: ≤12%

  • Solubility: Sparingly soluble in water

  • Microbial Limits: Conforms to pharmacopeial requirements

Applications in Pharmaceutical Formulation:

Used orally to treat iron deficiency anemia, particularly in patients with chronic kidney disease (CKD). It also serves as a phosphate binder to reduce serum phosphate levels in CKD patients on dialysis.

Comments:

Ferric citrate offers dual therapeutic benefits as both an iron supplement and a phosphate binder. Its good tolerability profile and mild gastrointestinal side effects make it suitable for long-term use in CKD patients. However, its relatively low iron bioavailability may require dose adjustment based on therapeutic need.
